Pectinase Production by Using Coffee Pulp Substrate as Carbon and Nitrogen Source

About thirty-five percent of coffee pulp waste is pectin. It may potentially be a source to used in the bioprocessing industry. For example, it can as substrate produce pectinase from microorganisms under solid-state fermentation (SSF). In this investigation, an isolated fungus VTM4 with density 10 7 spores/mL was grown on medium-based, and after 0-168 hours incubation at 30 °C, activity detected. The measured based reducing sugar released by crude against 0.5% alkali extract pectin 20 mM buffer acetate pH 5. highest produced 223.34 µg/mL 72 °C. optimum enzyme 4 maximum 0.747 U/mL stable (more than 80%) range 4-5.5. results revealed that could utilized carbon nitrogen pectinase. Further research purification characterization improve yield production needed.
